Laboratory centrifuge selection and classification

The centrifuge is a high-speed rotating machine with complicated structure. It is a special equipment for rapidly separating the mixed solution by using the centrifugal force and the difference in the sedimentation speed of different substances in the centrifugal field. It is a centrifuge tube containing the sample solution. A special device in which a bottle or a bag is placed on a centrifugal shaft and the centrifugal force of the rotor is rotated at a high speed around the shaft to separate particles of different properties in the sample. Analysis and separation of samples can be achieved.

Since the advent of the centrifuge, the progress of low speed, adjustment and overspeed has been mainly reflected in the design of centrifuge and centrifugal technology, and the two complement each other. Because the desktop centrifuge is simple in structure, low in cost, small in size, and soon becomes a routine laboratory instrument, almost all well-known centrifuge manufacturers at home and abroad produce desktop centrifuges, such as Germany's Hheraeus, eppendof, Sigma, which have been active in China in recent years. Hettich, Hermle, Jouran of France, Beckman of the United States, Sorvall, IEC, Hitachi, Kukna, Sanyo/Mse of Japan, Beijing Medical Centrifuge Factory, Hunan Kaida Scientific Instrument Co., Ltd., Shanghai Anting Scientific Instrument Factory, etc. . From the perspective of rotational speed, desktop centrifuges are basically in the category of low-speed, high-speed centrifuges, and therefore have the characteristics of low-speed, high-speed centrifuges. Compared to floor-standing centrifuges, they are only smaller in size and capacity.

There are many types and models of centrifuges, which are domestic and imported. According to the application, they can be divided into analytical centrifuges and preparative centrifuges. According to the speed, they can be divided into:

Ordinary centrifuge (low speed) <8000r/min;
High speed centrifuge 8000 ~ 30000r / rain;
Ultracentrifuge 30000 ~ 80000r / min;
Ultra high speed centrifuge >80000r/min;

Select the centrifuge according to the high molecular weight of the organism:

Molecular weight >10. , using ordinary centrifuges and high-speed centrifuges;
The molecular weight is 10 to 10. Using an ultracentrifuge;
Molecular weight <10, separation by centrifugation and ultracentrifugation is not good, and additional separation methods must be used.

According to the purpose of the experiment and the needs of the experiment, centrifuges with different capacities, different speeds and different temperature control can be selected. For example, when it is necessary to use normal temperature, micro, and rapid centrifugation, a palm type centrifuge (LX-100 type, manufactured by Jiangsu Haimen Kirin Medical Instrument Factory) and a micro centrifuge (DW-41 type, KOREA Korea) can be used; More than 16000 rpm, can use desktop centrifuge (Hettich, MIKRO12-24 type in Germany; do small amount of plasmid DNA extraction, cDNA extraction experiments, protein extraction, and extract RNA from plants, the rotation speed needs 12000 - 140000 rpm, often using high-speed frozen desktop centrifugation Machines (Hettich, 16R and 32R in Germany), benchtop high-speed centrifuges (Hettich MIKRO12-24 in Germany) and high-speed refrigerated centrifuges (JOURN BR4i in France); often used in the preparation of competent cells and large amounts of plasmid DNA Automatic high-speed freezing centrifuge (HITACHI, Japan, Hitachi Industrial Co., Ltd., Model 20PR-52D) and ultra-high-speed refrigerated centrifuge (Beckman AvantU-301, USA); if large-scale centrifugation is required, for example, 50mL, 100mL or more, and the rotation speed If you need 20,000 to 70,000 rpm, you can use the automatic ultra-speed freezing centrifuge (HITACHI, Japan Hitachi Industrial Co., Ltd., 70P) Type 72). Among the above centrifuges, the American Beckman Avanti J-301 ultra-high speed refrigerated centrifuge is superior in performance. It combines vacuum, high speed and cooling, and can be used for a large number of cells and sub-cells. Rapid separation of bacteria and bacteria is a common laboratory equipment. Centrifuges with a speed range of 5000 to 30000 RPM (revolutions per minute) are usually referred to as high-speed centrifuges. The maximum speed of Beckman Avanti J-301 centrifuges is 30000RPM, so it belongs to the range of high speed centrifuges.
