A new method of stem cell reprogramming - "indirect lineage conversion"

A new method for generating stem cells from mature cells is expected to increase the production of stem cells in the laboratory and help eliminate barriers to regenerative medicine.
A new technology developed by researchers at the Salk Institute for Biological Studies can get an unlimited amount of stem cells and their derivatives, while reducing production time by more than half, from nearly two months to 2 weeks. The results of the study have been published this week in Nature Methods.
One obstacle that needs to be overcome before stem cell therapy is widely used is how to produce enough stem cells at a rapid enough rate for acute clinical applications. Stem cells are highly valued for their pluripotency and have the ability to become almost all cells. There are two sources of stem cells for research and clinical applications: direct access to pluripotent stem cells, or reprogramming of mature cells into pluripotent stem cells.
Researchers have developed a reprogramming methodology called "indirect lineage conversion (ILC)": after a brief exposure to a reprogramming factor, the somatic cells are pushed back to a plastic intermediate state, followed by Differentiate again.
Using this method, the researchers successfully converted human fibroblasts into mesodermal progenitor cells, which have bidirectional differentiation potential and can differentiate into endothelial cells and smooth muscle cells. Specifically, the researchers transformed human fibroblasts into CD34+ progenitors with bidirectional differentiation potential. In vivo, these differentiated endothelial cells exhibit neo-angiogenesis and anastomasis.
This indirect lineage conversion methodology provides a simple and efficient technique. In this strategy, the complete process of reprogramming to pluripotency is shortened or bypassed, and the transformation of somatic cells undergoes an intermediate state of plasticity.
The study proved for the first time that this involves partial dedifferentiation (partial de-d generation).
