Preventive measures: 1 Use age-appropriate bacteria, bacteria age is 30 to 35 days, hypha vitality. 2 In high-temperature weather, arrange to inoculate in the morning or night. The culturing room should strengthen ventilation and reduce temperature, or delay the cultivation time appropriately to avoid high-temperature injury. 3 Prevent scalding hyphae during inoculation.

Second, germ seed germination does not eat material

In the normal germination environment, mycelia of mycelium germinated well after inoculation, and the color was white, but it did not grow in the material. Causes: 1 Use fresh wood chips or doped pine chips as raw materials, which contain substances that inhibit the growth of mycelium. 2 culture material water content is too high. 3Using urea to supplement nitrogen source, the amount is too large; or bacteriostatic antibacterial agent is added in excess to inhibit mycelial growth. 4 bagging too much, poor air permeability, lack of oxygen content. 5 Incomplete sterilization of culture materials, bacteria multiply. 6 The pH of the culture medium is either acidic or partially alkaline, making it difficult for the mycelium to grow. The quality of 7 strains was poor, their viability was declining, and their ability to eat mycelium was weakened.

Preventive measures: 1 Use old hardwood sawdust instead of fresh sawdust and pine wood chips. 2 Make up the nitrogen source with high-quality bran and rice bran, and add no less than 20% of the high-temperature time. It is best not to add corn flour at high temperatures. The addition of urea should not exceed 0.1%. It is best not to add it. The amount of carbendazim inhibitor is 0.05% to 0.1%, and it is better not to add it. (3) Adding a certain amount of corn cobs (pulverized) into wood chips will help improve the physical properties of the culture materials, solve the problems of excessively fine wood chips, small voids, and insufficient oxygen. 4 Master the correct sterilization operation. When sterilizing by high-pressure steam, the cold air must be exhausted. Sterilization should be ensured for 2 hours under the pressure of 147.1 kPa. When the pressure cooker is sterilized, the lid is tight and does not leak gas. The firepower must be prosperous and steam must be sufficient. The temperature is kept at 100°C for 10 hours or more. 5 When the material is bagged, the pH is adjusted to 7-8. 6 master the ratio of good material and water, culture material moisture content, yellow varieties controlled at 65%, white varieties of 60%, reduce the high temperature, do not over-wet. 7 To purchase seeds from reputable strain supply units, choose white, thick and dense quality strains of mycelium.

Third, mycelial yellow shrink

10-15 days after inoculation, the mycelium gradually yellowed, sparse, and atrophied and could not continue to grow in the material. Causes: (1) The room temperature is high, the ventilation is not good, and the bag and the bag are discharged too tightly, which affects the air circulation. The material temperature is difficult to distribute and the mycelium is damaged by high temperature. 2 The material is too wet and the pressure is too high, the air permeability is poor, and the mycelium is deficient in oxygen. 3 sterilization is not complete, the material within the proliferation of thermophilic bacteria, compete for nutrition, inhibit mycelial growth. 4 The ventilation in the culture room is not good, and the concentration of carbon dioxide is high.

Preventive measures: 1 It is better to keep the temperature of the culture room at about 20°C. Bags and bags should be slightly spaced so that the material temperature can be dispersed. High temperature weather, good ventilation and cooling. 2 grasp the ratio of good material. Bags are suitable for elasticity. When the material is found wet, the bag can be moved to a ventilated place. 3 The culture medium was sterilized at normal pressure and kept at 100°C for 10 hours to prevent bacterial contamination.

Fourth, the late mycelial growth is slow, delayed bag reasons: 1 bag airtight, mycelial hypoxia, more common in two closed-mouth bacteria culture. 2 The temperature is low, the mycelium grows slowly or stops growing.

Preventive measures: 1 The use of closed bacteria culture, when the mycelium into the material 3 to 5 cm long, the two ends of the bag tie rope untie, loose bag mouth, into the air, or the use of puncture ventilation oxygen supplement. 2 Maintain proper temperature culture, indoor temperature not lower than 18 °C.
